A formulated therapeutic oral agent consisting of a combination of 5-fluorouracil (5-FU) and uracil (1:4). Because of its higher concentration in the combination, uracil saturates the uracil-reducing enzymatic activity of dihydropyrimidine dehydrogenase, thereby inhibiting first-pass dihydropyrimidine dehydrogenase-mediated hepatic metabolism of the uracil analogue 5-FU and permitting oral administration of 5-FU. 5-FU inhibits the enzyme thymidylate synthase, resulting in decreased thymidine synthesis and so reduced DNA synthesis and tumor cell cytotoxicity.
