An orally bioavailable quinazoline with potential antineoplastic and radiosensitizing activities. CI-1033 binds to the intracellular domains of pan-erbB tyrosine kinases, irreversibly inhibiting their signal transduction functions and resulting in apoptosis and suppression of tumor cell proliferation. erbB is a member of the epidermal growth factor receptor (EGFR) family and is implicated in the malignant transformation of many solid tumors, including breast, ovarian, prostate, lung, and brain cancers.
